Description
α-Conotoxin BuIA is a peptide toxin comprising 13 amino acids, originally isolated from Conus bullatus. Notably, this toxin features an atypical 4/4 disulfide bond structure. Despite its unique conformation, it exhibits potent antagonistic activity at specific subtypes of nicotinic acetylcholine receptors (nAChRs), including α6/α3β2β3, α3β2, and α3β4 like some α4/7 conotoxins. In addition, α-conotoxin BuIA is able to kinetically distinguish between receptor subtypes containing either the β2 or β4 subunits. It achieves this by displaying fast off-rates for nAChRs with the β2 subunit, while maintaining very slow off-rates for those with the β4 subunit. Due to these properties, α-conotoxin BuIA serves as a valuable tool for differentiating between nAChRs with varying α and β subunit compositions.
