Description
α-Conotoxin EI is an 18-residue peptide derived from the venom of Conus ermineus, the only fish-hunting cone snail found in the Atlantic Ocean. This peptide exhibits a remarkable specificity for the nicotinic acetylcholine receptor (nAChR) in mammalian skeletal muscle as well as in the electric organ of Torpedo. Compared to other α-conotoxins, α-conotoxin EI demonstrates a novel selectivity profile. Notably, although it shares the same disulfide bond framework as the α4/7 conotoxins that target neuronal nicotinic acetylcholine receptors, α-conotoxin EI uniquely antagonizes the neuromuscular receptor, similar to the behavior of α3/5 and αA conotoxins. Its distinct binding preference for the α1/δ subunit interface of the Torpedo neuromuscular receptor renders α-conotoxin EI a valuable structural template for superimposing various α-conotoxins that exhibit their own receptor subtype specificities. This exceptional binding behavior not only highlights the versatility of α-conotoxin EI but also enhances our understanding of receptor interactions within the nAChR family.
