Description
α-Conotoxin GIC is a 16-residue peptide belonging to the α4/7-conotoxin subfamily that has been isolated from the venom of the cone snail Conus geographus. This remarkable toxin exhibits a potent inhibitory effect on the α3β2 subtype of the human nicotinic acetylcholine receptor (nAChR), demonstrating a pronounced selectivity for neuronal receptors over muscle-type receptors. Its high affinity for the human α3β2 subunit combination positions α-conotoxin GIC as the most neuronally selective nicotinic antagonist characterized to date. Despite sharing some sequence similarities with α-conotoxin MII, another potent antagonist of the α3β2 nicotinic receptor, α-conotoxin GIC is notably less hydrophobic. Furthermore, the kinetics of channel blockade exhibited by α-conotoxin GIC are significantly different from those of α-conotoxin MII, reflecting distinct mechanisms of action.
