Description
α-Conotoxin GID is a peptide that was originally isolated from the venom of the marine cone snail, Conus geographus. This peptide consists of 19 amino acids and adopts a specific folded structure stabilized by two disulfide bonds, which connect cysteine residues Cys1 to Cys3 and Cys2 to Cys4. Based on the amino acid composition between the second and third cysteine residues (referred to as loop I) and between the third and fourth cysteine residues (loop II), α-conotoxin GID is classified within the α4/7-conotoxin family. Notably, α-conotoxin GID exhibits selective inhibitory activity against neuronal nicotinic acetylcholine receptors (nAChRs). It is able to inhibit both α7 and α3β2 nAChRs and is at least 1000-fold less potent than the combination of α1β1γδ, α3β4, and α4β4. α-Conotoxin GID also potently inhibits the α4β2 subtype. What sets α-conotoxin GID apart as a unique member of the α4/7-conotoxin family is its ability to effectively block both the α7 and α3β2 isoforms, a characteristic that distinguishes it from other conotoxins such as PnIA and PnIB, which display a more selective, opposite binding profile. In addition, it has the potential to inhibit chronic pain.
