Description
α-Conotoxin LtIA is a peptide toxin comprised of 16 amino acids, has been isolated from the venom of the Conus litteratus. This toxin is part of the α-conotoxin superfamily, which is known for its ability to target and inhibit nicotinic acetylcholine receptors (nAChRs), integral membrane proteins involved in signal transmission in the nervous system. Previously characterized α-conotoxins have a highly conserved Ser-Xaa-Pro motif, which is essential for effective nAChR interaction. Despite lacking this highly conserved motif, α-conotoxin LtIA exhibits a significant inhibitory effect specifically on α3β2 nAChRs with an IC50 of 9.8 nM, providing an important molecular probe for studying the structure and function of α3β2 nAChR.
In contrast, even at ten times higher concentrations, α-conotoxin LtIA shows minimal to no inhibitory effect on α4/β2 and α3/β4 nAChRs. Notably, α-conotoxin LtIA exhibits promising analgesic properties through its specific interaction with the α3β2 nAChR subtype. The finding highlights its potential for future painkiller research and development.
