Description
α-Conotoxin PIA is a peptide toxin originally isolated from the Conus purpurascens. It acts as a selective antagonist for nicotinic acetylcholine receptors (nAChRs) containing the heteromeric α6 subunit. By binding to the extracellular domain of nAChRs, α-conotoxin PIA effectively distinguishes between the α6 and α3 subunits, exhibiting a lower affinity for the latter. Characteristic of the α4/7 conotoxin subfamily, α-conotoxin PIA displays an "ω-shaped" topology. However, unlike other neuron-targeting α4/7-conotoxins, it uniquely features a protruding N-terminal tail comprised of Arg1-Asp2-Pro3. This is due to the formation of a stable type I β-turn involving Asp2-Pro3-Cys4-Cys5. Furthermore, a kink induced by Pro15 within the toxin's second loop creates a distinct steric and electrostatic environment. Structural features in α-conotoxin PIA may be responsible for its unique receptor recognition characteristics. These unique properties and structural elements make α-conotoxin PIA a valuable tool for studying neurological conditions such as Parkinson's disease and schizophrenia.
