Description
αA-Conotoxin PIVA is both the first member of a new conopeptide family, αA-conotoxins, and the first representative of a new structural class of conopeptides, isolated from the venom of the cone snail Conus purpurascens. One of the most distinctive features of αA-conotoxin PIVA is its unique amino acid sequence and cysteine framework, which markedly differ from those of other nicotinic acetylcholine receptors (nAChRs)-targeted peptides previously identified in Conus venoms. Despite these structural differences, αA-conotoxin PIVA effectively blocks the acetylcholine binding site at the neuromuscular junction, mirroring the action of the more well-known α-conotoxins. It functions by reversibly blocking postsynaptic muscle nAChRs, specifically the fetal α1/β1/γ/δ configuration. In addition, it inhibits the adult α1/β1/ε/δ configuration of these receptors, albeit with a reduced affinity. αA-Conotoxin PIVA can be used to study neurological diseases, and its high receptor specificity highlights the diverse bioactive potential harbored by marine life.
