Description

Ciguatoxin 3C is a lipophilic neurotoxin characterized as a cyclic polyether compound featuring a rigid structure composed of 13 rings fused by ether bonds, known for its potent biological activity. This molecule is a secondary metabolite produced by certain species of microalgae, specifically from the genera Gambierdiscus and Fukuyoa. Additionally, ciguatoxin 3C can be found in the tissues of various marine invertebrates and fish native to the Pacific region. Notably, a significant aspect of ciguatoxin 3C is its ability to activate voltage-gated sodium channels, which can lead to a spectrum of neurological symptoms in affected individuals, including dysesthesia and paresthesia.

Mechanism of Action

Ciguatoxin 3C predominantly targets ion channels in neuronal membranes, where it exerts significant effects on voltage-gated sodium channels. By modulating these channels, Ciguatoxin 3C causes depolarization of the neuronal membranes, thereby affecting synaptic transmission. This toxin induces a rapid inward tonic current in neurons, mainly facilitated by gamma-aminobutyric acid (GABA). As a result, there is an enhancement of inhibitory postsynaptic currents, accompanied by a reduction in excitatory postsynaptic currents. This alteration of the excitatory-inhibitory balance may be responsible for several of the neurological symptoms observed in ciguatera poisoning.
