Description
ω-Conotoxin GVIA, a neurotoxin containing 27 amino acids and three disulfide bonds, was first isolated from the venom of the fish-hunting cone snail Conus geographus. It acts as an antagonist of voltage-activated N-type Ca2+ channels and neurotransmitter release at neuronal synapses, with its antagonistic activity attributed to a tyrosine residue at position 13. ω-Conotoxin GVIA is selective for N-type voltage-gated calcium channels and is approximately 40-fold more potent in vivo than morphine. Notably, it blocks specific voltage-dependent N-type Ca2+ channels in neurons but not in muscles. Furthermore, it has demonstrated antihypertensive, neuroprotective, and analgesic effects, making it of great interest in drug development.
